It happened at Barren Hill and River Roads in Whitemarsh around 1:30 Sunday morning.
Police say the driver of a Volkswagen Jetta lost control and slammed into a tree, trapping both the driver and her passenger inside for almost an hour.
They had to be cut free from the wreckage by firefighters before both victims were taken to waiting medical helicopters. One girl was flown to The Hospital of the University of Pennsylvania and the other was flown to Thomas Jefferson University Hospital.
The victims are hospitalized in critical condition.
Police say they are trying to determine if alcohol was involved.
