Police are now searching for the two suspects who then crashed into a SEPTA bus while fleeing the scene.
One of the attempted robbers was also hurt in an apparent shootout with the victim during the incident.
It happened at around 10 p.m. on Wednesday at S&S Candy Quick Stop at 40th and Brown streets.
Investigators said the tables were turned on a 17-year-old who was shot multiple times while he was allegedly trying to rob a man inside the store.
That teenager survived but the man, identified as 36-year-old Terrell Jones, of the 1200 block of North Ringgold Street, was shot and killed.
All of it was seen on surveillance video.
"You can see our initial shooting victim who died inside the store playing one of the video machines and that's when clearly two males enter the store, and it appears to be a robbery," said Chief Inspector Scott Small.
Small said there's then a struggle between the victim and suspects.
Police say one of the suspects fired a weapon at Jones, striking him. Jones then returned fire.
"It appears that the victim who died pulled his weapon and there was an exchange of gunfire," Small said.
Jones was found lying on the floor of the store with multiple gunshot wounds to the body. He was pronounced dead at the scene.
The two suspects jumped into a blue getaway car, described as a Mazda, driven by a third person, police said.
That car is then seen by city-owned cameras going the wrong way down Brown Street.
It crashed head-on into a SEPTA bus about a half block away, police said.
The three people inside the car ran from the scene, but police said they later found a 17-year-old nearby with gunshot wounds to the arm and chest.
They believe he was one of the suspects inside the store.
Police rushed him to hospital in stable condition, where he is being treated and held as a prisoner.
Investigators are still searching for two suspects who ran from the car.
The SEPTA driver and the eight passengers onboard were not injured, according to police.
A weapon was recovered and the investigation is ongoing.
