According to the Hollywood Reporter, the twins chose not to participate in Fuller House, though supported the upcoming Netflix show good wishes.

"Although Ashley and Mary-Kate will not be a part of Fuller House, I know how much Full House has meant to them and they are still very much considered family," executive producer Robert L. Boyett said in a statement. "It has been exciting to see how they have built their professional careers, and I support their choice to focus on their fashion brands and various business endeavors. I appreciate their support and good wishes towards Fuller House."
John Stamos, who returns to play Uncle Jesse, lamented the twins' decision on Twitter.

Fuller House will center around Candace Cameron Bure's character DJ Tanner. Now a 30-something widow raising three children, she will be joined by her sister Stephanie (Jodie Sweeten) and best friend Kimmy Gibler (Andrea Barber), who move in with DJ to help.
Stamos, a producer on the show, will also make appearances as Uncle Jesse, as will his on-screen wife Lori Loughlin as Aunt Becky, according to Cameron Bure. Bob Saget and Dave Coulier are reportedly still in negotiations, but are also said to make appearances as well.
