The accident was reported just before 11:00 p.m. Wednesday on the eastbound Turnpike near Bustleton Pike in Feasterville.
One of those injured was reported to be a pedestrian.
All four victims were taken to area hospitals.
There was no immediate no word on their conditions.
The multi-vehicle crash shut down two eastbound lanes for about two hours.
Pennsylvania State Police are investigating.
