The accident happened just before 12:30 a.m. Saturday at the intersection of Commons Boulevard and Airport Road.
A driver in a Jeep SUV with three people inside was travelling east on Airport Road and failed to handle a curve, police say.
The vehicle ran off the highway struck a tree, rolled over and landed in a ditch.
The impact of the crash trapped the victims inside the wreckage.
Police say 31-year-old Rhashem Revelle of Wilmington and 27-year-old Tynisha Scott of Bear were killed in the crash.
They had seat belts on but suffered severe head injuries.
Another victim, 26-year-old Andre Revelle of Wilmington was in the back seat.
He was taken to Christiana Hospital in critical condition.
The crash remains under investigation.
