It happened around 5:30 Saturday morning near mile marker 17.
Authorities say the victim, a man in his 50s, was in the car with his girlfriend when they got into an argument.
Police say the woman stabbed him once in the chest. He was taken to Hahnemann Hospital in critical condition.
The victim's girlfriend was treated at the University of Pennsylvania for minor injuries.
Pennsylvania State Police are investigating.
