The crash happened around 3:45 a.m. Sunday on Lombard Street near South 27th Street.
Police say the van was travelling westbound when the driver missed the curve in the road and went straight into a brick and metal fence.
Firefighters had to cut both men out of the van.
The victims were rushed to the hospital and were last reported to be in serious condition.
An investigation into the crash continues.
